Fluorescent Monitoring of Poly(lactic-co-glycolic Acid) Degradation and Extracellular Matrix Turnover of Engineered Tissue Constructs

Abstract
Development of fluorescent tagging techniques for inert polymers is an advantageous tool for monitoring biomaterial degradation online. Here we present a protocol for the fluorescent tagging of poly(lactic-co-glycolic acid) (PLGA) which allows for accurate monitoring of degradation. This was applied to PLGA with various lactic/glycolic acid ratios to determine cellular responses to degradation. The final aim was to produce a new metric, the turnover index (TI), which assesses cellular responses to biomaterial degradation.
